Application
Permeable supports, also known as cell culture inserts, are an essential tool for the study of both anchorage-dependent and independent cell lines.
This product produces a cell culture environment that closely resembles an in vivo state. Allow polarized cells to carry out metabolic activities in a more natural manner because the cells feed both apically and basolaterally.
Application: co-culture, epithelial cell polarity, migration, invasion, transport and permeability studies.
NEST Transfer Cap for High Efficiency Erlenmeyer Flasks
Two-Way Liquid Transfer Cap
The two-way transfer cap is used along with a 2L, 3L or 5L shake flask to
connect a liquid inlet tubing with the required device. The liquid transfer
is achieved by connection of a peristaltic pump between the shake flask and
the device. Upon completion of the transfer, the transfer cap can be
replaced with a vent cap for culture.
Multifunctional Liquid Transfer Cap
Unlike the two-way liquid transfer cap, the multifunctional transfer cap
can be directly placed in an incubator for culture after the liquid
transfer is completed. It can reach a large air flux. The sampling part is
composed of a sampling nozzle and a one-way valve, which can prevent the
liquid from flowing backwards during the sampling process and ensure the
aseptic sampling. The liquid inlet tubing is provided with a PTFE needle
filter, which solves the issue of liquid remaining in the tubing during the
feeding process.
Inverted Liquid Transfer Cap
The inverted transfer cap is used along with a 2L, 3L or 5L shake flask to
connect a liquid inlet tubing with the required device. When liquid
transfer is required, the liquid is transferred under gravity with the
inverted shake flask.
Application
It is applicable to liquid transfer and culture during mass proliferation of bacteria and suspension cells.
Product Features
- Closed transfer reduces the risks of contamination during liquid transfer
- The cap is molded and connected in an integrated manner, reducing the risks of leakage and media residues
- A variety of tubing diameters are available and aseptic welding of liquid inlet tubing under normal conditions is supported
- High-quality materials and smooth inner wall of the tubing provide an excellent transfer performance
- Sterility level (SAL) is maintained at 10-6
- Endotoxin-free, and no ingredients of animal origin

General Guidelines
Follow these guidelines when using ig 5-alpha chemically competent cells.
- Handle competent cells gently as they are highly sensitive to changes in temperature or mechanical lysis caused by pipetting.
- Thaw competent cells on ice, and transform cells immediately following thawing. After adding DNA, mix by tapping the tube gently. Do not mix cells by pipetting or vortexing.
Protocol
- Remove competent cells from the -80 °C freezer and thaw completely on wet ice (10-15 minutes).
- Aliquot 1-5 µl (1 pg-100 ng) of DNA to the chilled microcentrifuge tubes on ice.
- When the cells are thawed, add 50 μl of cells to each DNA tube on ice and mix gently by tapping 4-5 times. For the pUC19 control, add 1 µl of (10 pg/µl) DNA to a chilled microcentrifuge tube, prior to adding 50 µl of cells. Mix well by tapping. 4) Incubate the cells with DNA on ice for 30 minutes.
- After 30 minute ice incubation, heat shock the cells at 42 °C for 45 seconds.
- Transfer the tubes to ice for 2 minutes.
- Add 950 µl of Recovery Medium or any other medium of choice to each tube.
- Incubate tubes at 37 °C for 1 hour at 210 rpm.
- Spread 50 μl to 200 μl from each transformation on Pre-warmed selection plates.
- Incubate the plates overnight at 37 °C.
