GelNest™ Matrix

 

GelNest™ Matrix is derived from mouse tumor tissue and contains extracellular matrix components such as laminin, type IV collagen, heparan sulfate proteoglycans, and more. These components support cell adhesion, differentiation, and proliferation, providing signals for these processes. Additionally, they simulate the characteristics of the basement membrane in the physiological environment, enhancing the success rate and effectiveness of cell culture.

 

In addition to the matrix components, GelNest™ Matrix is rich in various growth factors. These growth factors promote cell differentiation, proliferation, and migration, mimicking cellular signaling pathways and interactions in the physiological environment. GelNest™ Matrix has a wide range of applications, particularly in tissue engineering, cell culture, and research. It can be used for organoid culture, stem cell differentiation, angiogenesis, migration or invasion assays, and in vivo tumor studies.

 


Storage and Operation Instructions

 

To store GelNest™ Matrix, it is recommended to keep it in a -20°C freezer until ready for use. Prior to initial use, thaw the gel and divide it into single-use portions. Store these portions in a -80°C freezer for a maximum of 2 years. Please note that GelNest™ Matrix Gel should not be stored in a frost-free freezer.

 

GelNest™ Matrix is in a liquid state at 4°C and transitions into a gel state at 37°C. It starts to solidify into a gel when the temperature exceeds 10°C. For optimal performance, it is advised to pre-chill the pipette tips and conduct the procedures on ice.

Erlenmeyer Flasks


MTC Bio’s Erlenmeyer Shaker Flasks are made of polycarbonate, PETG or styrene/butadiene (Jumbo Flasks only). The polypropylene screw caps are vented with 0.2µm membranes to increase air transmission and enhance cell growth. The mouth size conforms with the international SBS standard, for quick, secure connection to bioreactor equipment.
Polycarbonate flasks and Jumbo Fernbach flasks have baffled (ribbed) bottoms to provide increased oxygenation and mixing efficiency even at lower shaking speeds. Attention

  • During the cultivation process, the liquid should be taken through the gap between the upper layer and the hole. When adding liquid, be careful and slow to avoid the damage of the membrane;
  • Incubation of the permeable support in the medium prior to culturing the cells can improve cell attachment and distribution.


Guidelines

  • Pre-equilibration: After added culture solution to the pores of the multi-well plate and inserted the insert, please placed it in the incubator for at least one hour or overnight.
  • Fix and stain cells directly in the nest, and use a scalpel to cut the membrane for long-term storage. Before using please add culture solution to the pores of the multi-well plate, then insert the insert and add the culture solution which included cells to the insert.

  • Pre-equilibration: After added culture solution to the pores of the multi-well plate and inserted the insert, please placed it in the incubator for at least one hour or overnight.

NEST Transfer Cap for High Efficiency Erlenmeyer Flasks

Two-Way Liquid Transfer Cap
The two-way transfer cap is used along with a 2L, 3L or 5L shake flask to connect a liquid inlet tubing with the required device. The liquid transfer is achieved by connection of a peristaltic pump between the shake flask and the device. Upon completion of the transfer, the transfer cap can be replaced with a vent cap for culture.


Multifunctional Liquid Transfer Cap

Unlike the two-way liquid transfer cap, the multifunctional transfer cap can be directly placed in an incubator for culture after the liquid transfer is completed. It can reach a large air flux. The sampling part is composed of a sampling nozzle and a one-way valve, which can prevent the liquid from flowing backwards during the sampling process and ensure the aseptic sampling. The liquid inlet tubing is provided with a PTFE needle filter, which solves the issue of liquid remaining in the tubing during the feeding process.


Inverted Liquid Transfer Cap

The inverted transfer cap is used along with a 2L, 3L or 5L shake flask to connect a liquid inlet tubing with the required device. When liquid transfer is required, the liquid is transferred under gravity with the inverted shake flask.

It is applicable to liquid transfer and culture during mass proliferation of bacteria and suspension cells.

  • Aliquot 1-5 µl (1 pg-100 ng) of DNA to the chilled microcentrifuge tubes on ice.
  • When the cells are thawed, add 50 μl of cells to each DNA tube on ice and mix gently by tapping 4-5 times. For the pUC19 control, add 1 µl of (10 pg/µl)
  • DNA to a chilled microcentrifuge tube, prior to adding 50 µl of cells. Mix well by tapping.
  • Incubate the cells with DNA on ice for 30 minutes.
  • After 30 minute ice incubation, heat shock the cells at 42 °C for 45 seconds.
  • Transfer the tubes to ice for 2 minutes.
  • Add 950 µl of Recovery Medium or any other medium of choice to each tube.
  • Incubate tubes at 37 °C for 1 hour at 210 rpm.
  • Spread 50 μl to 200 μl from each transformation on pre-warmed selection plates.
  • Incubate the plates overnight at 37 °C.

ReadyStrain II™is the only product of its kind, providing all necessary parts for filtration of cell suspensions and combining them into a single, preassembled, ready to use, sterile packet. These kits eliminate the contamination risks associated with opening multiple packages and assembling separate pieces (i.e. tubes, strainers and caps). The individual sterile packets mean researchers are no longer forced to expose entire bags of 50mL tubes and caps in order to perform one filtration. Instead, they can open a single packet directly under the hood.

Each kit contains a sterile SureStrain™Strainer, pre-attached to a sterile 50mL tube, with a separate sterile screw-cap ready for assembly, processing and storage after the filtration is complete. Kits are supplied in easy open “tear-strip” bags and gamma sterilized.
