ION Biosciences Essentials Red Calcium

Red Calcium Essentials provides the necessary reagents for multi-well plate-based, fluorescence microscopy applications and high-throughput measurements of intracellular calcium (Ca²+) mediated changes through a wide variety of plasma membrane and intracellular calcium channels and transporters. Red Calcium Essentials is also useful for investigating numerous effectors of ion channels and transporters, including G-protein coupled receptors.

Our red calcium dye, ION Calcium Red-1 (ICR-1), is a red fluorescent indicator (Ex/Em: 580/660 nm) responsive to intracellular Ca²+ dynamics. ICR-1’s spectral properties allows for multiplexing with an indicator of another color or GFP-tagged cells.

When following the recommended protocol, Red Calcium Essentials provides enough reagents to make 100 mL of working solution, enough for ten 96-well plates. The actual number of assays will vary according to optimal dye concentrations for your application.

ION Biosciences IPG-1 TMA Salt+

Yellow-green fluorescent, potassium (K+) indicator, membrane

ION Potassium Green – 1 (IPG-1) is the best in class yellow-green fluorescent, intracellular potassium (K+) indicator for measuring changes at most intracellular K+ concentrations. IPG-1 has Ex/Em: 525 nm/545 nm and a high-sensitivity to detect small changes in K+ concentration. IPG-1 has a lower affinity (Kd = 50 mM) than IPG-2 (Kd = 18 mM) or IPG-4 (Kd = 7 mM).

Ratiometric calcium (Ca²+) indicator, membrane impermeable.
 
Fura-2 is the most popular UV-excitable, ratiometric green indicator for calcium (Ca²+) measurements. Ex/Em: 340/505 nm can be used to measure Ca²+-bound Fura-2, and Ex/Em: 380/505 nm can be used to detect Ca²+-free Fura-2. Ratiometry is optimal for imaging applications where quantification of intracellular Ca²+ concentrations is desired, and reduces effects of photobleaching, heterogenous dye loading, and variable cell morphology.

ION Biosciences ING-2 TMA+ Salt

Yellow-green fluorescent, sodium (Na+) indicator, membrane impermeable. Higher Na+ affinity than ING-1.
 
ION Natrium Green – 2 (ING-2) is a yellow-green fluorescent, intracellular sodium (Na+) indicator with improved cellular loading and significantly higher brightness than SBFI. Its spectral properties (Ex/Em: 525 nm/545 nm) and large dynamic range make ING-2 the best Na+ indicator for high-throughput screening applications targeting Na+ channels, and non-selective monovalent cation channels. ING-2 has a more physiologically relevant affinity (Kd = 20 mM) than ING-1 (Kd = 92 mM). Also compatible with fluorescence microscopy using common fluorescein, GFP or more ideally YFP filters.

Fluo-3 is a green fluorescent, intracellular calcium (Ca²+) indicator with slightly red-shifted spectral properties compared to Fluo-4. Offers a large dynamic range that can be better suited for high contrast imaging applications.Is preferred over Fluo-4 for some cell types.

ION Biosciences ICR-1 K+ Salt
 
ION Calcium Red – 1 (ICR-1) is a red fluorescent (Ex/Em 580nm/660nm) calcium (Ca²) indicator for intracellular Ca²+ measurements. ICR-1’s long-wavelength emission and a large Stokes shift reduces contributions of autofluorescence, making ICR-1 AM optimal for cellular and tissue imaging applications. ICR-1 can also be multiplexed with GFP-labeled cells or other green fluorophores commonly used in experiments. Unlike some other red fluorescent Ca²+ indicators, ICR-1 does not accumulate in the mitochondria. Data supporting fluorescence lifetime imaging and multiphoton imaging capabilities have also been demonstrated.

Fluo-2 is a green fluorescent, calcium (Ca²+) indicator. Fluo-2 has higher Ca²+ affinity (Kd = 290 nM), superior cell loading and comparable spectral properties compared to Fluo-4. Offers superior sensitivity versus other Fluo analogues at lower Ca²+ concentrations.
