ION Vital – Viability kit can be used to quantify live and dead cells within eukaryotic cell suspensions or adherent cultures, 3D cultures, organoids, and some non-fixed tissues. Cannot be used for yeast or bacteria. When following our protocol, ION Vital—Viability kit provides enough reagents to make 150 mL of working solution, enough for fifteen 96-well plates or 1,500 flow cytometry samples. The actual number of assays will vary according to optimal dye concentrations for your application.
ION Biosciences PBFI K+ Salt
 
PBFI is a UV-excitable, ratiometric green indicator for intracellular potassium (K+) measurements. Ex/Em: 340/505 nm can be used to measure K+-bound PBFI, and Ex/Em: 380/505 nm can be used to detect K+-free PBFI. It is 1.5X more selective for K+ over Naand works well in cytosolic conditions where [K+]/[Na] is ~10. Ratiometry is optimal for imaging applications where quantification of intracellular K+ concentrations is desired and reduces effects of photobleaching, heterogenous dye loading, and variable cell morphology.

ION Biosciences IPG-1 TMA Salt+

Yellow-green fluorescent, potassium (K+) indicator, membrane
 
impermeable. Lower K+ affinity than IPG-2 and IPG-4.
 
ION Potassium Green – 1 (IPG-1) is the best in class yellow-green fluorescent, intracellular potassium (K+) indicator for measuring changes at most intracellular K+ concentrations. IPG-1 has Ex/Em: 525 nm/545 nm and a high-sensitivity to detect small changes in K+ concentration. IPG-1 has a lower affinity (Kd = 50 mM) than IPG-2 (Kd = 18 mM) or IPG-4 (Kd = 7 mM).

Fluo-2 is a green fluorescent, intracellular calcium (Ca²+) indicator. Fluo-2 has higher Ca²+ affinity (Kd = 290 nM), superior cell loading and comparable spectral properties compared to Fluo-4. Offers superior sensitivity versus other Fluo analogues at lower Ca²+ concentrations.
Brilliant Sodium Assays
 
ION’s Brilliant Sodium Assay is a total assay solution for multi-well plate-based, high-throughput measurements of changes in intracellular Na+ mediated through a wide-variety of plasma membrane and intracellular sodium channels and transporters. In multi-well, plate-based formats, the Brilliant Sodium Assay can be used to discover and characterize the effects of many tens-of-thousands of compounds and environmental factors on effectors of intracellular Na+. ION’s Brilliant Sodium Assay provides all the reagents necessary for use as a wash or no-wash assay with adherent or non-adherent cells. The optional use of a probenecid solution and an extracellular background masking solution (TRS) offers the ultimate in compatibility for cells types which are difficult to load with fluorescent Na+ indicators (e.g. Chinese Hamster Ovary, CHO cells) and when performing assays in complete, serum-containing cell culture medium is desired.

ION Natrium Green – 2 (ING-2) is a yellow-green fluorescent, intracellular sodium (Na+) indicator with improved cellular loading and significantly higher brightness than SBFI. Its spectral properties (Ex/Em: 525 nm/545 nm) and large dynamic range make ING-2 the best Na+ indicator for high-throughput screening applications targeting Na+ channels, and non-selective monovalent cation channels. ING-2 has a more physiologically relevant affinity (Kd = 20 mM) than ING-1 (Kd = 92 mM). Also compatible with fluorescence microscopy using common fluorescein, GFP or more ideally YFP filters.
ION Biosciences Fura-2 LR AM
 
Leakage-resistant, ratiometric calcium (Ca²+) indicator, membrane permeable.
 
UV-excitable, ratiometric green indicator for intracellular calcium (Ca²+) measurements with longer cellular retention compared to Fura-2 AM and similar spectral properties. Ex/Em: 340/505 nm can be used to measure Ca²+-bound Fura-2, and Ex/Em: 380/505 nm can be used to detect Ca²+-free Fura-2. ION Biosciences ICR-1 AM

Red fluorescent, calcium (Ca²+) indicator, membrane permeable. Optimal for multiplexing and tissue imaging. Fluorescence lifetime and multiphoton compatible.
 
ION Calcium Red – 1 (ICR-1) is a red fluorescent (Ex/Em 580nm/660nm) calcium (Ca²+) indicator for intracellular Ca²+ measurements.

ICR-1’s long-wavelength emission and a large Stokes shift reduces contributions of autofluorescence, making ICR-1 AM optimal for cellular and tissue imaging applications. ICR-1 can also be multiplexed with GFP-labeled cells or other green fluorophores commonly used in experiments. Unlike some other red fluorescent Ca²+ indicators, ICR-1 does not accumulate in the mitochondria. Data supporting fluorescence lifetime imaging and multiphoton imaging capabilities have also been demonstrated.

Calcein AM
 
Green fluorescent, live cell indicator.
 
Calcein AM is a membrane-permeant, non-fluorescent form of calcein that is converted to green fluorescent calcein in viable cells, resulting in uniform cytosolic fluorescence (Ex/Em 495 nm/515 nm). Calcein is well retained within the cytosol of most healthy cells with intact cell membranes.
