Suggested Protocol using Taq 2x Mix

This protocol serves as a guideline for primer extensions. Optimal reaction conditions such as incubation times, temperatures, and amount of template DNA may vary and must be individually determined.


Notes:

Set up reaction mixtures in an area separate from that used for DNA preparation or product analysis. Work on ice at all times.

The final MgCl2 concentration of this 2x Master Mix is 1.5 mM. If more than 1.5 mM MgCl2 is required, use 25 mM MgCl2 (may be purchased separately) to adjust the Mg2+

 Thaw Taq 2 Master Mix and primers. It is important to thaw the solutions completely and mix thoroughly before use to avoid localized concentrations of salt. Keep all components on ice.

 

 The table below shows the reaction set up for a final volume of 50 m If desired, the reaction size may be scaled down. Use 10 µl of the Taq 2x Master Mix in a final volume of 20 µl.

 

 Important: Spin Taq Master Mix vials briefly before use.

Mix gently by pipetting the solution up and down a few times.

Program the thermal cycler according to the manufacturer´s instructions. For maximum yield and specificity, temperatures and cycling times should be optimized for each new template target or primer pair.

Place the tubes in the thermal cycler and start the reaction.

 Initial denaturation step. Optional, but recommended for genomic DNA

b Denaturation step: Heating the DNA template disrupts the hydrogen bonds yielding ssDNA.

c Annealing step: Primers anneal to ssDNA template. Typically annealing temperature is 3-5°C below the Tm of the primers.

d Extension/elongation step: Taq DNA polymerase synthesizes a new DNA strand complementary to the DNA template. Extension time depends on length of DNA fragment to be amplified. At optimum temperature the DNA polymerase will polymerize 1000 bases per minute.

 

e Final elongation step: After the last PCR cycle to ensure that any remaining ssDNA is fully extended.

The Accuris ASPIRE Laboratory Aspirator incorporates a quiet, maintenance-free pump inside a sturdy, yet compact base. The collection bottle incorporates a reliable Overflow Protection Valve that closes off the tube to the vacuum in the event of a full bottle. In addition, the replaceable in-line filter protects the vacuum pump from contaminants and any moisture that comes through the bottle. Graduations on the collection bottle indicate the amount of fluid collected and when it's ready to be emptied. The Handheld Vacuum Pipette has an ergonomic design that's comfortable in the hand during prolonged use.  Pressing the button applies the vacuum. The pipette accepts Pasteur pipettes, serological pipettes, and also comes with 2 accessory pipette cones. One cone is designed to accommodate 200/300ul pipette tips, the other has a thin, stainless steel capillary for fine control aspirating.
