Frequently Asked Questions:

Specifications:

Will these rockers work with different voltages (in different countries)?

Yes. The rocker uses 24V DC supplied by a small external power supply that plugs into a wall outlet. Next Advance will supply the appropriate power supply for the local power and wall outlet configuration.

What is the maximum load?

The rocker can handle 11 lbs. (5 kg).

What is the range for the tilt angle?

The amount that the rocker platform tilts back and forth can be adjusted from as little as +/- 1 degree to as much as +/- 18 degrees.

What is the difference between Infinity Rockers and Next Rockers?

Infinity Rockers and Next Rockers are both rugged, quiet, rockers with lots of power. Infinity Rockers have digital speed control and a digital display of the rocking rate in cycles per minute (cpm). Because Infinity Rocker motors are brushless, i.e. do not have brushes that can wear out, they will last for many years.

What is the rocking rate?

For Infinity Rockers, the rocking rate can be as slow as barely moving to a maximum speed of 80 cycles per minute (160 rocking motions per minute). Next Rockers are available in a regular speed range (4 to 80 cycles per minute) or super slow (1 to 20 cycles per minute).

What are the Super Slow versions?

The Next Rockers come in standard and super slow versions. For those applications that require slow, smooth rocking, we recommend the super slow (up to about 20 cycles per minute) versions. They provide finer speed control at the lower speeds. The motor in these versions is geared down for slower operation and greater power, just as gears on a bicycle allow for more power at lower speeds.

Can I use these rockers in incubators or cold rooms?

Our rockers can operate in incubators (up to 60°C) or cold rooms (down to 4°C) if the temperature is kept constant and the humidity is not excessive. Temperature swings which cause condensation will damage the electronics and void the warranty. In fact, the electronic circuitry includes additional heat sinks to ensure reliable operation at elevated temperatures.

Operation:

How do I adjust the tilt angle?

To adjust the angle, use a flat blade screwdriver to loosen the labeled screw on the rear of the instrument, slide the linkage to the desired position, then tighten the screw.

How do I adjust the rocking rate?

You can turn the knob on the front of the instrument to adjust the rocking speed.

Can I clean the rubber mat?

Yes. You can remove the rubber mat from the platform and clean it with soap and water. Let it dry before placing it back on the platform.

VitroGel® Angiogenesis Assay HC Kit

  • Xeno-free functional hydrogel system supporting angiogenesis process
  • Control and study the effect of the hydrogel mechanical properties on the angiogenesis process
  • Multiple applications in one kit:  Tube formation, invasion, and animal injection
  • Control the hydrogel properties:  Add your own growth factors and compare with positive control
  • Harvest cells within 20 minutes at 37 °C without enzyme solution

VitroGel® Angiogenesis Assay Kit is a revolutionary tool for researchers to study the effect of both hydrogel properties and culture medium on angiogenesis process. The kit can be used to study the angiogenesis tube formation and invasion on both 2D hydrogel coating and 3D cell culture methods. The VitroGel® system is also good for animal injection for in vivo studies.

Angiogenesis is a highly regulated process that involves the growth of new blood vessels from the existing vasculature. This process plays an important role in both normal developmental processes and numerous pathologies, including wound healing, tumor growth, and metastasis to inflammation and ocular disease.

Traditional angiogenesis assay highly relies on natural extracellular matrix (natural ECM), which has non-adjustable hydrogel compositions and properties.  Therefore, our understanding of the angiogenesis process is limited by studying the molecular cues such as growth factors and inhibitors in culture medium only. There is a lack of knowledge on how the properties of hydrogel affect the angiogenesis process.

There are two versions of VitroGel® Angiogenesis Assay Kits:

  • VitroGel® Angiogenesis Assay Kit (Cat No. VHM06-K):  Ready-to-use with a fixed hydrogel mechanical strength to support the angiogenesis assay with adjustable supplements.
  • VitroGel® Angiogenesis Assay HC Kit (Cat No. TWG011-K):  Assay kit with a tunable high concentration hydrogel to allow full control of the hydrogel’s mechanical strength with adjustable supplements.

Depending on the kit type, the tunable VitroGel Angiogenesis Assay HC Kit can contain the following components:

  • VitroGel® AAK-HC, a tunable, xeno-free high concentration hydrogel.
  • AAK Dilution Solution, for adjusting the hydrogel concentration
  • AAK Supplement 1, a hydrogel supplement without vascular endothelial growth factors (VEGFs) for promoting cell attachment and growth.
  • AAK Supplement 2, a hydrogel supplement with VEGFs to promote tube formation and as a positive control.
  • All the components can be purchased separately.

Besides molecular cues, the VitroGel® Angiogenesis Assay HC Kit allows researchers to explore the effects of hydrogel mechanical properties on angiogenesis.  The high concentration VitroGel® AAK-HC hydrogel is room temperature stable and can be adjusted by simply mixing the hydrogel solution and dilution solution at different rations (recommend 1:1 to 1:5 v/v) to achieve different mechanical strengths. The diluted hydrogel solution can be directly mixed with supplement at 2:1 (v/v) ratio for hydrogel formation. Researchers can adjust the molecular cues of the hydrogel by adding the growth actors/inhibitors directly to the supplement before mixing with VitroGel® AAK-HC.  Cells cultured in this system can be further harvested easily with the VitroGel® Cell Recovery Solution.

Cyto3D™ Live-Dead Assay Kit

Live dead cell viability analysis for 3D and 2D cell culture.

The Cyto3D™ Live-Dead Assay Kit is used to determine the live/dead nucleated cells by using a fast one-step staining procedure for analysis on a dual-fluorescence system. This kit is recommended for viability analysis of cells cultured in 3D, 2D coating and on monolayer.

  • Ready-to-use
  • Fast
  • Sensitive
  • Excellent for 3D cell cultures
  • Cost-effective

Acridine orange (AO) and propidium iodide (PI), both nuclear staining (nucleic acid binding) dyes, are used in this kit. AO is permeable to both live and dead cells and stains all nucleated cells to generate green fluorescence. PI only penetrates the membranes of nucleated cells with compromised membranes and stains the dead cells to generate red fluorescence. Due to the quenching, when cells are stained with both AO and PI, all live nucleated cells fluoresce green and all dead nucleated cells fluoresce red (the PI reduces the fluorescence intensity of the AO by fluorescence resonance energy transfer (FRET)). Non-nucleated materials such as red blood cells, platelets and debris do not fluorescence and are ignored by fluorescence microscopes.

Dual-Fluorescence Viability, using AO and PI, is the recommended viability analysis method for cell lines, primary cells, and stem cells.

VitroGel® ORGANOID

ready-to-use, xeno-free (animal origin-free) hydrogel system for organoid culture

  • Xeno-free hydrogel for organoid formation
  • Supports a wide range of organoids from patient-derived samples, stem cells, tissues, co-culture and PDX resources
  • Scalable protocol for lab automation
  • Easy 20-minute cell harvesting (enzyme-free)
  • VitroGel® ORGANOID (1-4) are xeno-free (animal origin-free) hydrogels that support the growth of patient-derived organoids or organoids developed from pluripotent stem cells (PSCs), co-culture, and PDX model.

VitroGel® ORGANOID hydrogels are ready to use at room temperature and have a neutral pH, transparent, permeable, and compatible with different imaging systems. The solution transforms into a hydrogel matrix by simply mixing with the cell culture medium. VitroGel® ORGANOID hydrogels are good for both 3D cell culture and 2D hydrogel coating applications.

The VitroGel® ORGANOID hydrogels can work in conjunction with VitroGel® STEM (a xeno-free hydrogel for 3D static suspension cultures and scale-up of human pluripotent stem cells) by transferring stem cell spheroids cultured in VitroGel® STEM to VitroGel® ORGANOID hydrogels for organoid differentiation. See Xeno-free 3D Organoid Workflow B.  Key growth factors and molecules can be directly mixed with the hydrogel matrix or by adding on the top of the hydrogel. Organoids cultured in this system can be easily harvested out with our VitroGel® Cell Recovery Solution.  VitroGel® ORGANOID hydrogels provide a well-defined 3D microenvironment for the future of personalized medicine.

Which organoid hydrogel to choose

We still strongly suggest starting with the VitroGel® ORGANOID Discovery Kit (Item # VHM04-K) to make a quick screening to find the hydrogel version that is optimal for the organoid conditions.

The kit includes all four types of organoid hydrogels:

VitroGel® ORGANOID-1, VitroGel® ORGANOID-2, VitroGel® ORGANOID-3, VitroGel® ORGANOID-4)

Each of the four types has various bio-functional ligands, mechanical strengths, and degradability to fulfill the needs of different organoid culture conditions.

Because there are a wide range of organoid cell resources (from stem cells, patient-derived tissue, co-culture, to PDX), it is hard to tell which organoid hydrogel would best perform the researcher’s experiment. Therefore, the Discovery Kit helps researchers to screen the four different formulations to determine the best organoid hydrogel version for the organoid conditions.

From our findings, versions 1, 2, or 3 are suitable for gastric organoids. Versions 1 or 3 are suitable for lung organoids. Versions 2 or 3 are suitable for brain organoids, and versions 3 or 4 are suitable for cancer organoids.
