Pro Tip: Colored plastics do not have any impact on DNA amplification.  But when setting up qPCR it is often recommended to use white plastics.  This will limit or prevent fluorescence refraction.  Light refraction can minimize sensitivity and consistency. 

Pro Tip 2: If you are seeing inconsistencies in PCR/qPCR try PR1MA Pipette Tips.  This will help optimize and reduce variance in the reactions.  Simple test, see the residual liquid left in tips?  This means some of the taq, dNTPs, sample, etc is left in the tip.  PR1MA tips help reduce that significantly and improve your consistency. 

Pro Tip 3: Seeing evaporation in the outer wells of a PCR/qPCR plate?  Sometimes it’s the plate, sometimes it the seal, and sometimes it’s the cycler.  Call us we have solutions.

Accuris SmartBlue Transilluminators

 

Accuris SmartBlue Transilluminators have been developed for viewing DNA gels stained with new, safe fluorescent dyes.  These new dyes, such as SmartGlow, SYBR Green, Gel Green, have an excitation wavelength of 465-470 nm (visible blue light), and are safer to use than Ethidium Bromide.

Safer for samples, safer for you: Accuris SmartBlue Transilluminators are the perfect partner for the most common safe DNA stains. These instruments emit blue light at a wavelength of 465 nm, perfect for excitation of most green fluorophors, including our Accuris’ SmartGlow, SYBR Green, SYBR Safe, SYBR Gold, GelStar, GelGreen, to name a few.  SmartBlue transilluminators can also work for excitation of EtBr (though fluorescence is not as bright as with UV), see our Technical Bulletin with sample images.   Exposing DNA to blue wavelength does not cause molecular damage, and results in a much higher downstream cloning efficiency as compared to even short UV light exposures.  In addition, the visible blue light is not damaging to the user’s skin and is less harmful to the eyes.*


Adjustable filter cover: Both the E4101 and SB4500 transilluminators come with emission filter covers. These amber covers, which rest above the viewing surface and gel, filter out the blue light, allowing the fluorescing DNA bands to be seen clearly.  The covers can be set to a flat position for general gel viewing or can be set at an angle to provide easy access to the gel for band excision.  The SB4500 has a two-position filter cover, and the E4101 features a hinged filter cover.

 

Optional amber viewing glasses: Our E4000-VG1 SmartBlue Plus Viewing Glasses offer efficient blocking of the blue light wavelength from the transilluminators, allowing clear visualization of the fluorescing DNA bands without the filter cover in place.  These convenient glasses are recommended for users that are frequently excising DNA bands from gels.  SmartBlue Plus glasses also block 100% of UV wavelength so they can also be used for viewing gels on a UV transilluminator.

 

UI technology, bright and clear images: The bright LED light plates and filter layer provides an extremely evenly lit viewing surface. Even in ambient light, bands of DNA can clearly be seen in stained gels. Images obtained using the SmartBlue Transilluminator are brighter and more balanced than those obtained using epi-illuminators which light from the sides.


Reliable, durable, and long-lasting: An array of super bright LEDs with a long, 30,000 hour service life provide the light source for the SmartBlue
 Transilluminators. Unlike those in a UV transilluminator, the filters in the SmartBlue systems will not solarize and degrade in performance over time. Gel bands can be excised directly on the scratch resistant, glass viewing surfaces. To save energy, the power switch includes an automatic 5 minute shutoff. SmartBlue Transilluminators are covered by a 2 year warranty.

 

* SmartBlue products use high intensity LED lights.  Although the visible blue wavelength is significantly safer than UV light, it is recommended to not stare directly at the viewing surface without the amber cover in place.

DNA SafeStain is a new and safe nucleic acid stain for visualization of double-stranded DNA, single-stranded DNA, and RNA in agarose gels. The dyes are developed to replace toxic ethidium bromide (EthBr, a potent mutagen), commonly used in gel electrophoresis for visualization of nucleic acids in agarose gels. DNA SafeStain is non-carcinogenic by the Ames-test. The results are negative in both the mouse marrow chromophilous erthrocyte micronucleus and mouse spermary spermatocyte chromosomal aberration tests.

DNA SafeStain emits green fluorescence when bound to DNA. It has two excitation wavelength peaks when bound to nucleic acid, at 290nm and 490nm and the emission can be detected at 530nm.

Storage:
Store under dark at 4°C or room temperature.

User Instruction: 
DNA SafeStain can be used in the exact same way as EtBr in agarose gel electrophoresis. For example, add 100µl to 1 liter of 1X running buffer (1xTAE or 1x TBE), and use this DNA SafeStain containing buffer to make the agarose gel and run the gel.

 

Special Notes: 
1. For best results, dilute the DNA SafeStain 1:10,000 in your gel running buffer and use this dye-containing running buffer to make your agarose gel and to run the gel.

2. As is the case with EtBr; add more DNA SafeStain, if brighter DNA bands are desired. To view the results, any conventional DNA gel viewing light box or gel documentation system should work well with the DNA SafeStain; although, using a LED light instead of a UV light source is preferred due to the harmful nature of UV light.

DNA SafeStain Plus is a highly sensitive green fluorescent DNA/RNA staining reagent for detecting nucleic acids in agarose and polyacrylamide gels. This unique stain gives high sensitivity for detection of double-stranded or single-stranded DNA and RNA. Gels can be post-stained or the stain can be added to gels during gel casting or to the gel running buffer. DNA SafeStain Plus has two excitation wavelength peaks at about 290nm and 490nm, and an emission wavelength at 530nm, making it compatible with a standard UV light box, a blue-light transilluminator, or a gel reader equipped with visible light excitation; such as, a 488 nm laser-based gel scanner.

Users only need to add templates and primers, and water if needed. Extra 25 mM magnesium solution is also provided for additional fine adjustments. The Red Mixes contain a non-hazardous inert red dye. The purple mixes contain a non-hazardous inert purple dye. Completed PCR reactions utilizing the D124-R (red dye) or D124-P (purple dye) can be directly loaded into a well of agarose gel or other gels, for electrophoresis. No extra loading buffer is needed.

Storage: 4°C for up to one month, or -20°C for long term storage.

Magnesium Chloride: In general, 1.5 mM MgCl2 is recommended; this may vary with different conditions and primer sets. Some primers/templates may require adjustments for MgCl2 concentration, which can be achieved as shown below:

Directions for use: For a 50 uL reaction, use 25 uL of the Taq Master Mix, add template, primers, and water to a final volume of 50 uL. Cycling conditions vary for different templates and primers. To start with, try 30 cycles as follows: denature at 94°C for 30 seconds, anneal around 55°C for 30 seconds, and extend at 72°C for 1 minute / kb. After the PCR cycles, extend at 72°C for another five minutes to complete the PCR. Then store the reaction at 4°C.

1x Composition: 10 mM KCl, 20 mM Tris HCl (pH 9.0), 16 mM (NH4)2SO4, 0.1% Triton X-100, 1.5 mM MgCl2, 200 mM dNTPs, 2.5 units / 25 uL of Taq DNA polymerase, (optional: trace amount of red or purple dye) and enzyme stabilizers.
