The OD600 DiluPhotometer™ is a small, easy to use instrument that is dedicated to measuring samples near the 600 nm wavelength. It is suitable for measuring growth rates of several cell types including E. coli and yeast. Use of the OD600 DiluPhotometer™ with Implen's DiluCell™ 10 facilitating the measurement of low volume samples (volume requirement of 200 µL and 100 µL respectively). The  DiluCell™ 10  allows for automatic 1/10 and 1/20 dilutions. With a bandwidth of 40 nm, the OD600 is also suitable for protein quantification assays like Bradford (595 nm). The small size and portable nature of the OD600 DiluPhotometer™ make it convenient for use in incubation cabinets and under anaerobic conditions. 

The OD600 DiluPhotometer™ has been designed for the spectrophotometric analysis of bacterial and yeast growth rates and the quantification of proteins at a wavelength nearby 600 nm (e.g. Bradford). The instrument is battery operated and can be easily transported for usage in incubation cabinets and under anaerobic conditions. The batteries last for almost 1 month when fully charged. A 600nm LED source in combination with a fiber optic is used to obtain the optical density measurement. Spills can be easily wiped from the surface, and the cell compartment area can be rinsed with, formaldehyde or ethylene oxide to sterilize it.

Lambert-Beer-Law

The Lambert-Beer-Law, also known as Beer's Law, empirically relates the absorption of light to the properties of the sample. This law states that there is a logarithmic relationship between the transmission of light through a specific sample (T = I/Io with I = outgoing light and Io = incoming light), the molar extinction coefficient for a specific compound (ε), the concentration of the absorbing species in the material (c) and the distance the light travels (d). By leaving all conditions constant but changing the pathlength (distance the light travels), you can bend the Beer-Law to get an automatic dilution of your sample. This principle is realized within the OD600 DiluPhotometer with DiluCell.

Automatic Sample Dilution

Previously it was necessary to use standard cuvettes with a volume of at least 1 ml and additionally to dilute the sample manually. DiluCell from Implen allows for lower sample volume requirements and provides automatic virtual sample dilution. Due to the unique design of DiluCell cuvettes, the light-path is reduced from the standard 10 mm to 0.5 mm (DC 20) or 1.0 mm (DC 10). According to the Lambert Beer Law this shortened pathlength results in an automatic sample dilution by factor 20 (DC 20) or 10 (DC 10). Automatic dilution with DiluCell saves time and excludes dilution errors and cross contaminations.

Measuring Cell Density

The stage of growth of a bacterial culture needs to be monitored to ensure that the cells are harvested at the optimum point (towards the end of log phase) for the greatest density of live cells. The optical density of the sample indicates when this point has been reached. This value varies dependent on the cells being grown. Routinely cells are grown until the absorbance at 600 nm, also known as OD600, reaches approximately 0.4 prior to induction or harvesting. A linear relationship exists between cell number (density) and OD 600 up to approx. 0.6.

The Accuris SmartDrop™ series of nano spectrophotometers provides quick and easy quantification of small samples. Utilizing a small sample volume eliminates the need for dilutions, providing more accurate results.

A full color, 7” touchscreen with icon-driven menus and preprogrammed protocols gets users up and running quickly. No external computer is required. Each unit features a quartz pedestal onto which a drop of sample is placed. The arm is lowered and the drop is stretched into a liquid column of specificed length for spectral analysis. DNA, RNA and protein concentrations and purity numbers are displayed in a matter of seconds.

The resulting data can be printed, saved in the SmartDrop’s onboard memory or exported via USB as an Excel file. All SmartDrop™ models include an integrated printer. For OD600 readings, each model includes a cuvette port and two optical glass cuvettes. Three SmartDrop™ models are available to meet a range of applications and budgets.

SmartDrop™ L is the affordable choice for laboratories needing to determine nucleic acid and protein concentrations and purity of samples. The LED light source and silicon photocell sensor provide readings at 260 and 280nm. Samples from 1.0 to 2.0 µL are loaded directly on the pedestal. Results are provided in about 5 seconds. The software automatically calculates and displays 260/280 ratios.

A full spectrum UV-Vis spectrophotometer, SmartDrop™ X requires 0.5 to 2.0 µL of sample. In addition to nucleic acid and protein calculations, the software has preset programs for BCA, Bradford and Lowry assays. A full spectrum analysis can be performed in about 6 seconds, aiding in the identification of unknowns. Up to 5 specific points on the spectrum can be selected for colorimetric readings output. The long life Xenon lamp source requires no warm up and is paried with a highly sensitive CCD detector. To provide the most accurate results, the SmartDrop™ X utilizes 2 path lengths.

This top of the line spectrophotometer can perform not only UV-Vis readings in the range of 200-800nm but also fluorescence readings. Like the SmartDrop™ X, the SmartDrop™ XF features predefined programs for nucleic acids, proteins, and various colorimetric assays. The patented, motorized system gently stretches the sample column and allows for three different path lengths, providing accurate readings of samples from 0.5 to 2.0 µL without dilution. The stable optics and two detectors measure your samples and reference simultaneously  providing superior measurement accuracy. 

The variable spectral bandwidth provides your experiments with robust detection and quantitation limits, as well as optimal data precision and repeatability. This flexible platform allows the user to comply with multiple regulatory agencies using one spectrophotometer- saving money and time. Three scan speeds are available with wavelength scan intervals of 0.1, 0.2, 1, 2 or 5 nm. The large sample chamber is outfitted with a 10 x 10 mm cuvette holder, but allows for an 8-position cuvette changer (order 83070-61 separately) for flexibility.

The integrated user interface provides local controls with the touch of a keypad. The colored graphical display is easy to read and will display spectrum as well as kinetic curves. Prism PC software is supplied with the spectrophotometer to assist the user with post-measurement analysis. The Prism PC software can be used to fully control the spectrophotometer and comes pre-loaded with methods for DNA analysis, including 260/280 and 260/230 ratios with 320 nm correction. Under the multi-wavelength measurement mode, up to 20 wavelengths can be measured simultaneously and quantitation curves can be created by measuring up to 200 standards.
